چکیده

1 Micro and Nanotechnology Laboratory, 2 Institute for Genomic Biology, 3 Beckman Institute for Advanced Science and Technology, University of Illinois, Urbana –Champaign, IL, 61801 USA ABSTRACT We have created a randomly distributed nanocone substrate on silicon and coated with silver for surface-plasmonenhanced fluorescence detection and cell imaging. We observed three dimensional fluorescence enhancement on our substrate evidenced from the confocal fluorescence imaging of Chinese Hamster Ovary (CHO) cells and transfected HeLa cells grown on the substrate. The fluorescence intensity was amplified more than 100 fold as compared to glass substrate. Strong scattering within nanostructured area coupled with random total internal reflection inside the cell resulted in the observed enhancement in fluorescence further away from the substrate surface.

Enhanced live cell imaging via photonic crystal enhanced fluorescence microscopy.

We demonstrate photonic crystal enhanced fluorescence (PCEF) microscopy as a surface-specific fluorescence imaging technique to study the adhesion of live cells by visualizing variations in cell-substrate gap distance. This approach utilizes a photonic crystal surface incorporated into a standard microscope slide as the substrate for cell adhesion, and a microscope integrated with a custom illu...

Engineering of core/shell nanoparticles surface plasmon for increasing of light penetration depth in tissue (modeling and analysis)

Objectives: In this article, a new procedure for increasing the light penetration depth in a tissue is studied and simulated. It has been reported that the most important problem in biomedical optical imaging relates to the light penetration depth, and so this makes a dramatic restriction on its applications. In the optical imaging method, the detection of the backscattered pho...

Imaging of Surfaces by Concurrent Surface Plasmon Resonance and Surface Plasmon Resonance-Enhanced Fluorescence

Surface plasmon resonance imaging and surface plasmon induced fluorescent are sensitive tools for surface analysis. However, existing instruments in this area have provided limited capability for concurrent detection, and may be large and expensive. We demonstrate a highly cost-effective system capable of concurrent surface plasmon resonance microscopy (SPRM) and surface plasmon resonance-enhan...

Dual-Color Fluorescence Imaging of EpCAM and EGFR in Breast Cancer Cells with a Bull’s Eye-Type Plasmonic Chip

Surface plasmon field-enhanced fluorescence microscopic observation of a live breast cancer cell was performed with a plasmonic chip. Two cell lines, MDA-MB-231 and Michigan Cancer Foundation-7 (MCF-7), were selected as breast cancer cells, with two kinds of membrane protein, epithelial cell adhesion molecule (EpCAM) and epidermal growth factor receptor (EGFR), observed in both cells. The membr...

Surface Plasmon Resonance Sensors on Raman and Fluorescence Spectroscopy

The performance of chemical reactions has been enhanced immensely with surface plasmon resonance (SPR)-based sensors. In this review, the principle and application of SPR sensors are introduced and summarized thoroughly. We introduce the mechanism of the SPR sensors and present a thorough summary about the optical design, including the substrate and excitation modes of the surface plasmons. Add...
